I have made a booking on wotif.com for a hotel this evening. However, my partner had tried to book direct and was told "no availability". Should I be worried?

You're probably ok -- many hotel booking sites "buy" blocks of rooms from hotels to then sell on their own sites. So, from the hotels perspective, they are "sold out", even though some of the rooms may still be available for booking on various sites.
However -- the best thing to do now is call the hotel back directly and confirm that they have your reservation in their system. If they don't, contact wotif.com IMMEDIATELY and have them make sure they send your res to the hotel. If not, then you can panic.

I have used wotif.com many times and always happy with the service.
But of course, that is NOT a guarantee that EVERYONE have ha similar experience. But I have not heard negative experience form anyone about this site yet.

As others have said, WOTIF seems to work fine.
I have used them many times in various countries with no problems.
This of course is no guarantee that it will work for you

I've never had any problems with wotif and I've used them many times.
Why not call the place you're staying to confirm your booking, that's what I always do.
